There is a very extensive and specific set of instructions for setup in the stickie posts at the top of this forum. It will answer questions you haven't asked yet.

I always run 2,3 and sometimes 4 filters on a new setup for the first 24 hrs after adding sand and water. Just add filter floss to the compartment and nothing else, you should see a difference within 24 hrs.

Wait and give folks a chance to respond...its not instant. Is it sand? If so let it settle before turning on the filter.
